Course Details

Covers the use of high performance liquid chromatographic techniques in chemical analysis. The course starts with a brief review of separation theory applied to liquid chromatographic separations. Subsequent sections of the course cover topics related to more advanced aspects of high performance liquid chromatography. Topics include maintenance of liquid chromatographs, optimizing high performance liquid chromatographic separations, preparative scale liquid chromatography and validation in liquid chromatography. Laboratory exercises provide practical training of the principles covered in the lectures.


CHEM 5000 or CHEM 6609 or an equivalent introductory analytical chemistry course.

Gravitying Outcomes

Upon successful completion of this course, the student will be able to:

  • o Demonstrate a thorough knowledge of the theory of liquid chromatography.
  • o Explain the general steps involved in the operation of liquid chromatographic systems.
  • o Select the correct logical steps involved in liquid chromatography troubleshooting.
  • o Demonstrate procedures for the maintenance of the following liquid chromatographic components.
    • o Pumps including binary and quaternary
    • o Sample injection systems including autosamplers
    • o Columns including guard columns and analytical columns
    • o Detectors including UV/Visible, Diode Array, Fluorescence
    • o Valves
  • o Select operating parameters for a liquid chromatographic separation by choosing appropriate mobile phase composition, column type and detector.
  • o Demonstrate a thorough knowledge of chiral separations in liquid chromatography.
  • o Assess the suitability of pre-column or post-column derivatisation techniques for a range of liquid chromatography applications.
  • o Develop strategies for quantitative and qualitative analysis in LC using common software packages.
  • o Explain the general steps involved in preparative scale liquid chromatography.
  • o Demonstrate a thorough knowledge of instrumentation validation in liquid chromatography.
